Squire G and Ralph W Grayson were indeed William's sons, but by his first wife, unknown. As you point out, the Montgomery Co. TN marriage record for William Grayson and Susannah Tinnin was dated 8 Sep 1802, but Squire G Grayson was born 3 Mar 1799 in Christian Co. KY (d. 1871 Freestone Co. TX), and Ralph W. Grayson was born about 1800 in Montgomery Co. TN and died 1850/1860 in Sumter Co. AL, where he was listed as age 50 in the 1850 Census.
I did not know about William and Susannah having a daughter Mary Grayson, but you may be right. The widow Susanna Grayson's 1830 census record in Wayne Co. MS showed her with two daughters, one age 5-10 (b. 1820/1825) and the other 15-20 (b. 1810/1815).We both show that Amanda Melvina was one daughter, although I have no details on her.
There was a Mary A. Grayson, b. c1826,who m. Dr. Samuel E. Casteel 22 Dec 1849 in Sumter Co. AL.But there seems little doubt that she was daughter of Ralph Grayson, who m. Catherine McCarty 16 Oct. 1824 in Marengo Co. AL.
