My grandmother, who died a few years past, is Helen Gibson Tuttle the sister of your descendant, Florence, listed in the 1900 census. My grandmother confirmed before she died that her father was John Jackson Tuttle and John's father was Edmund Tuttle, both from Rockaway, NJ. My grandmother was born and raised in Rockaway. John J. and "Edward" H. are listed in the Morris Co., Rockaway, NJ 1897-1898 village directory. The directory lists their homes on Maple Avenue. I suspect that Edmund also went by the name "Edward." John's brother, George E. is also listed in the city directory.
As you've noted, census records show that Samuel L. Tuttle is the father of Edward H., Rosina M., and Emma L. Tuttle. Samuel's wife was Mary Elizabeth Zindle, who is buried in the Rockaway Presbyterian Cemetary. Rockaway cemetary records indicate that Samuel L. and Mary E. also had a daughter Sarah R. Tuttle. The fact that Sarah was born in 1850, died in 1869, and a "Sarah" is not listed in the 1860 census but Rosina is, and Rosina is not listed in the 1870 census, makes me believe that Sarah R. (Rosina?) and Rosina M. may be the same person. Joseph Zindle, who appears in the Samuel L. Tuttle household in the 1860 census, may be a younger brother or nephew of Mary E.
I have not been able to determine the parents of Samuel L. Tuttle from my limited research to date. Alva Tuttle, author of "Tuttle-Tuthill lines in America," lists Samuel L. Tuttle, but does not give his parent's names. Alva indicates Samuel L. died in 1881 and is buried next to his wife Mary E. I have not confirmed this information, but Alva indicates the information was obtained from tombstone inscriptions. If we could determine the middle names of Edmund H., Rosina M. and Emma L., this might shed some light on Samuel's parents if the middle initial is also a family name. For example, if the middle initial for Edmund "H" stands for Hopkins, then this would link Samuel L. to parents Samuel Tuttle and Lydia Hopkins of Sussex Co., Hardyston Twp., NJ, as you've suggested. If the "H" stands for "Hall" then this is Mary E.'s mother's maiden name - Lydia Hall. Lydia married Charles Zindle.
Another possible link is Henry D. Tuttle and Joseph F. Tuttle, who are also listed in the 1897-98 Rockaway village directory. Henry and Joseph are in the 1880 census records as father and son, respectively. Henry (born abt. 1828) is about 4 years younger than Samuel L. (b. 1824) based on their ages reported in the census. This could imply that they are brothers. No other Tuttles appear in the 1897-98 village directory.
Let me know what you've learned.
