December 1794 in Anne Arundel County, Maryland. He married Susanna Lusby on July 11, 1786 in Anne Arundel County, Maryland134, daughter of Robert Lusby and Deborah Lee.

James Mayo died young before thirty years of age.

James Mayo was listed in the 1790 census as having 2 males above 16, 1 male less than 16, 1 female and 5 slaves. His brother, John Mayo, made him the executor of his will and left him farm animals ( Anne Arundel Co. Intestates liber JG1, folio 66). His father, Joseph Mayo, 1st, in his will ( Anne Arundel Co. Wills liber TG1, folio 309) left part of his land described as part of Selby's Marsh as follows: "begin the division line at the north end of the NW by N line being the first line of the whole tract and running thence across the said tract to the head of the easternmost branch of the cove called the Spring Cove where the main Road down the neck melded with the shore. All that part of Selby's Marsh lying on the southeast side of the division and on the northeast and east of the aforesaid road to belong to my son, James."
