Being a Jew is certain spiritual state.Within the original family of Jews coming from Abraham, Jacob and Isaac, so it is possible to induce a genetic relationship but this is only within a certain probability as Jews have intermarried with converts over the centuries.
Rachav (Joshuas wife), Moses wife Tzipora (Daughter of the famous convert Jethro), Ruth (King Davids grandmother) to name a few.

In the end genemapping can only strongly suggest a family relationship which one can then follow up on.It is the papertrail afterall as we all know that makes the historical difference in the end.

But in the end, because a persons mother is a Jew or if that person was converted by a rabbinical court then the person is Jewish as this endows on a person a certain spiritual state.
